Taken from NGC's coverage of the game based on their experiences at this year's E3. Some slight spoilers are present for the opening of the game so if you don't want to know, don't read any further.
The opening
Link starts the game as a shepherd outside of a small village called Toaru. The village is attacked by a group of bandits, who kidnap two of the locals (the mayor's daughter, Ilia, and a boy named Colin). This and the unlocking of a twilight dimension are what cause Link to take up the sword. Link then pursues the bandits across Hyrule on horseback, ending in a horseback duel with the bandit chieftain.
The Wolf 'gimmick'
Link transforms into a wolf upon crossing over into the twilight world, which is a complete mirror of Hyrule. Initially imprisoned, Link is freed by a character called Midna. Midna rides on Link's back whenever he's in wolf form, using her scarf to pick up items, and isn't the Twilight Princess. Although Link is weaker while being a wolf, he gains the ability to speak with animals and sniff out special items.
Other information? Twilight Princess is based on the Wind Waker game engine, which is still evident in the camera movement and combat (sword strikes freeze the game momentarily as you build up combos for example). Also added in is a new finishing move for weaker enemies if you knock them down (from the screenshots it looks like Link's downward impaling move from Smash Bros. and the GC version of Soul Calibur 2). There's a Forest Temple which is crawling with helper monkeys (and a few unhelpful ones) and a rather large Boss, resembling a mutated, slavering Deku plant with an eyestalk and huge teeth .
This is shaping up to be really impressive. Eiji Aonuma (the producer) is promising Twilight Princess will outdo Ocarina in every way imaginable, and if they can pull it off it'll be a fantastic game.
