Wolfy: The 14th Amendment allowed individual freedoms to trump states' rights through its definition of citizenship and a bit of judicial interpretation. Previously a state could infringe on Constitutionally-guaranteed rights: the Bill of Rights only limited the federal government. Now most of the rights and freedoms granted in the first ten amendments (but particularly the first) are protected from both federal and state infringement, largely because of various court cases. Practically, this means freedom of religion is protected in any government action, not just Congressional lawmaking.

I'm not sure about the 10th: Firefox will probably elucidate.
