So if you've ever played Warcraft 3 multiplayer, specifically DotA then you know how it plays. It's super easy to learn but each champion has a different skillset and some are harder than others to master. It's almost entirely mouse driven, though like most games proficient players hotkey things to their keyboards.
In my experience most games take between 30 minutes to an hour tops to finish. Unless the opposing team is playing serious defense, which can make it take forever.
The game seems to have a pretty intelligent matching system too so you don't get paired up with people who are way more experienced than you. This is done by having a global "summoner level" that is separated from your champion's level in the game. Champion levels reset with each match, so the faster you level the more likely you are to win.
You can purchase items and powerups from a vendor at your base as you go along and gain gold from kills and bonuses.
