The Lizard is in it, or rather Doctor Connors. It remains to be seen just how much of the Lizard will be in the movie. The real villain is most likely a character they are calling Van Atter. If you go back through Spider-Man lore, Van Adder (as it was spelled in the comics) was an employee of OSCORP and was one of the early test subjects for the goblin formula. Things went bad and not only did it affect his mind but it also mutated him, giving him claws, fangs, thick skin that can withstand some bullet fire, and glowing eyes. He was known as the Proto-Goblin. He only appeared in one comic.
This would make sense, since redoing the origin story sort of requires the Goblin as the first villain and they've already done Norman as the Green Goblin. This doesn't make sense, however, in the timeline because the Proto-Goblin came way before Peter Parker was ever Spider-Man. Still, I think they're taking artistic liberties and the Proto-Goblin will be the first real Villain. The first movie will build the relationship between Parker and Connors which will lend itself to a sequel in which Spider-Man finally has to square off against the Lizard to save Doc Connors.
